Major Mio Sakamoto, callsign "Samurai," serves as sub-commander of the 501st Joint Fighter Wing. She stands as the first Strike Witch, pioneering the Striker Unit technology created by her acquaintance, Dr. Ichiro Miyafuji. Born August 26, 1924, in Fuso (Japan analog), she is the oldest member of the 501st, initially 19 and later 20 during her service.

Her early combat against the Neuroi, including participation in the 1937 Fuso Sea Incident as a 1st Class Flight Petty Officer, forged her into a seasoned warrior. This experience defines her role as a strict mentor responsible for training recruits. While generally amicable, she can become overly assertive. An enchanted "magic eye" concealed beneath her eyepatch grants enhanced visual acuity to pinpoint Neuroi cores, a trait inspired by historical ace Sakai Saburo who also lost an eye.

Physically, Mio stands 165 cm (5'5") tall with black hair typically in a ponytail and olive eyes. She wields the sword Reppumaru/Reppuzan, employing specialized techniques like Zen Reppuzan and Reppu Genkotsu in combat. Her Striker Units draw from historical Fuso aircraft designs. Although her advanced age diminishes her combat shield strength, she retains exceptional flight proficiency and marksmanship.

Personally, Mio has a daughter, Nori Sakamoto, and descendants Mei Akiyama and Kiyoko Akiyama. Her familial connections extend into later timelines where she is noted as immortal post-1970. She possesses knowledge of the Way of Tea but doesn't strictly follow formal etiquette. A notable vulnerability is her fear of critters, especially snakes, which caused her to faint when stranded.
