Ayunosuke Odago, child of Koinosuke Odago, is charged with continuing his mission to find a worthy successor for the Lovely Eyepatch should he fail. Her appearance and name lead others to initially mistake her for a boy, despite being female. After Koinosuke's spiritual departure, Ayunosuke emerges as the new bearer of the duty, following 300 years alone in the wilderness awaiting her purpose. This long isolation results in a disheveled appearance and limited speech ability, reflecting her feral upbringing.

Ayunosuke actively pursues Jiyu Nanohana to deliver the Lovely Eyepatch, persisting despite Jiyu's rejection of the role. Her dedication mirrors her father's, though she demonstrates greater competence in ninja skills like evasion and healing. When Jiyu firmly renounces the eyepatch and declares her unwillingness to fight, Ayunosuke physically transforms, aging into her 300-year-old form from exhaustion and despair. This condition reverses only when Jiyu temporarily resumes her duties as Yagyu Jubei.

During the conflict with the Siberian Yagyu clan and Freesia Yagyu, Ayunosuke becomes a target due to her guardianship of the hidden eyepatch, relying on evasion for survival. Later, a spiritual manifestation of Koinosuke appears to guide her to the afterlife. Ayunosuke ultimately chooses to return to the mortal world and is shown residing in her father's former doghouse, indicating her continued presence alongside Jiyu.
