The anime adapts several of Marple's original cases: "Strange Jest," "Tape-Measure Murder," "The Case of the Perfect Maid," "The Blue Geranium," and "4:50 from Paddington." These adaptations retain the core plots and settings while compressing them for the episode format. Minor adjustments occur for cultural or linguistic reasons, such as altering a clue in "Strange Jest" that originally relied on an English idiom; the clue instead references a stamp depicting a winking angel. Such changes do not fundamentally alter Marple's deductive process or the mysteries' resolutions.

Marple collaborates indirectly with Poirot through Mabel, who bridges their separate investigative worlds, facilitating the adaptation of stories involving both detectives. Marple's character remains consistent with her portrayal in the original novels: she never married, has no immediate family, and relies on her nephew Raymond West for occasional financial support. Her background includes a childhood in a cathedral close and education at an Italian finishing school.
