Born with a disfigured left foot that didn't impede his movement, his childhood discovery of a fraternal twin brother, Domenico, who died shortly after birth, fostered deep contemplation on fate and inequality, shaping his path toward priesthood. At fifteen, laboring in an ossuary, he encountered DIO, who bestowed upon him a fragment of a Stand-bestowing Arrow and healed his foot.
A year later, substituting for an absent priest, Pucci heard a woman confess she swapped her deceased infant with Domenico, proving his brother lived. Meanwhile, his sister Perla dated Wes Bluemarine, later confirmed as Domenico. Bound by confessional secrecy, Pucci couldn't reveal their kinship. To halt the unknowingly incestuous relationship, he hired a private investigator group—unbeknownst to him, Ku Klux Klan members. They lynched Wes for his partial African American heritage and assaulted Perla. Wes survived, but Perla, believing him dead, died by suicide. This trauma awakened Pucci's Stand, Whitesnake, capable of extracting memories and Stands as DISCs. Confronting Wes, Pucci revealed their relationship and used Whitesnake to erase Wes's memories to spare him pain, leading Wes to adopt the name Weather Report.

As chaplain of Green Dolphin Street Prison, Pucci framed Jotaro's daughter, Jolyne Cujoh, for manslaughter to lure Jotaro. During a prison visit, Whitesnake stole Jotaro's memory and Stand DISCs, leaving him comatose.
Pucci deployed multiple Stand users as assassins against Jolyne and her allies, including Ermes Costello, Foo Fighters, and Emporio Alniño—a child whose mother Pucci had killed. He also manipulated inmates using DISCs, implanting the Stand Marilyn Manson into Miraschon. Discovering DIO's bone was vital for the heaven plan, Pucci had Sports Maxx reanimate it, creating the Green Baby. Fusing with the Green Baby at Cape Canaveral transformed Whitesnake into C-MOON, which inverted gravity around its influence point. Replicating new moon gravitational conditions evolved C-MOON into Made in Heaven, exponentially accelerating time.
