Colonel MacDougall is the primary antagonist in the anime film Spriggan, serving as the leader of the U.S. Machine Corps, a rogue military faction that has broken away from the Pentagon. He speaks with a commanding authority, as demonstrated when he declares the Ark's excavation site to be holy ground, directly challenging the protagonist, Yu Ominae. His motivations are not rooted in simple malice but in a cold, calculated agenda to seize control of the Ark's power, which includes the ability to control weather and manipulate genetic structures, and use it to reshape or destroy the world as he sees fit.

Within the story, MacDougall's role is to act as the primary obstacle and final threat that the Spriggans, Yu Ominae and Jean-Jacques Mondo, must overcome. He leads the assault on the ARCAM dig site at Mount Ararat, taking control of the Ark and holding its lead scientist hostage. His relationship with the protagonists is purely adversarial, defined by a brutal clash of ideals. However, his physical state is one of constant precariousness. The source of his power is also the source of his fragility; the immense psionic energy he wields threatens to tear his own body apart, forcing him to rely on his cybernetic implants and other technological means to prevent self-destruction. This weakness becomes a crucial factor in the climax of the film.
