Aoi Tohsaka, born Aoi Zenjou, is the wife of Tokiomi Tohsaka and the mother of Rin Tohsaka and Sakura Matou in the story of Fate/Zero. She comes from the Zenjou family, a bloodline that, while no longer practicing magecraft by her generation, carries a unique and valuable magical factor in its genetics. It was specifically for this latent quality in her bloodline that Tokiomi sought her hand in marriage, a union that produced two daughters with exceptional magical potential.

In terms of personality, Aoi is portrayed as a quiet, gentle, and traditionally composed woman, giving the impression of a well-bred young lady who has become a mother. She is described as a peaceful mother figure, in contrast to the more dynamic Irisviel von Einzbern. Her defining trait is her unwavering, almost blind loyalty to her husband and her complete acceptance of the values and traditions of the magus world. She understood that Tokiomi held an inhuman set of values as a magus, yet she supported him unconditionally, demonstrating a personal conviction that some observers note contains its own form of profound distortion. She is described as being careful, diligent, and dedicated to maintaining her household, prioritizing her duty to the Tohsaka family above all personal feelings.

Aoi's primary motivation is to fulfill her role as the wife of a magus. She made the conscious decision when marrying into the Tohsaka family to abide by its customs, no matter how severe. This conviction is most powerfully demonstrated when she consents to the adoption of her second daughter, Sakura, to the Matou family, a decision she accepts as a necessary obligation of a magus lineage despite the personal pain it causes her. Her deep love for her children is genuine, but it is always secondary to the demands of her husband's world.

Within the story of the Fourth Holy Grail War, Aoi serves as a key supporting figure. Her primary narrative function is as the object of affection for Tokiomi’s rival, Kariya Matou, whose desperate wish to save her from her unhappy fate drives his actions throughout the war. She has no magical abilities herself, possessing no Magic Circuits, though her awareness of magecraft is greater than that of a typical civilian. A notable moment in the war occurs when she returns to Fuyuki City in a panic after her daughter Rin runs away, showing her vulnerability as a mother outside the protected sphere of her home.

Her most significant and tragic relationships are with the three key men in her life. With her husband, Tokiomi, she shares a relationship based on duty and admiration, described as being a huge fan of his cool father persona. With Kariya Matou, she shares a past as a childhood friend who acted as an older sister figure to him, and she remains largely unaware of the depth of his unrequited love for her. Finally, her relationship with the priest Kotomine Kirei is one of trust, as she considers him an acquaintance due to his association with her husband.

Aoi's personal development is ultimately a tragic descent. At the war's climax, she discovers her husband’s body with a mentally and physically broken Kariya standing nearby. Misled into believing Kariya is the murderer, she lashes out in grief, accusing him of never having loved anyone. This psychological blow, delivered at his most fragile moment, causes Kariya to snap and strangle her. She survives the attack but suffers severe brain damage from oxygen deprivation, leaving her mentally incapacitated and bound to a wheelchair. In this state, she remains unaware of her husband's death or her daughter's absence. She survives for several years after the war but passes away before the start of the Fifth Holy Grail War, her fate standing as one of the conflict's most devastating civilian casualties.
