Description
Nurse Makiho Amano is a key supporting character in the 2000 anime film Blood: The Last Vampire. She serves as a quiet foil to the film's protagonist, Saya, representing ordinary humanity in the face of terrifying supernatural events. Her full name is Makiho Caroline Amano. Within the story, which is set in 1966, Makiho works as the school nurse at the high school located on the American Yokota Air Base.

Makiho is initially portrayed as a friendly, somewhat meek, and unassuming individual who gets along well with the students. Despite being apparently American, she speaks fluent Japanese. Her position as a school nurse, however, does not command much respect from figures like the school's principal, indicating her status is relatively low within the school's hierarchy. She is described as being plump, and her personality is gentle and kind, presenting a stark contrast to the dark and aggressive Saya.

Her primary role in the narrative is as an unwitting witness who becomes entangled in Saya's mission. At the school's annual Halloween party, two students who are actually chiropterans, bat-like vampire creatures, visit Makiho in the nurse's office. Saya suddenly arrives and attacks them, revealing their true monstrous nature. This traumatic event throws the gentle nurse into a state of profound shock.

Despite her initial terror, which is so extreme that she briefly contemplates suicide, Makiho demonstrates a crucial moment of development and inner strength. When Saya is trapped and needs assistance, Makiho is urged to help and finds the resolve to break down a set of locked doors, actively participating in her own survival and Saya's fight. Her motivations are simple and relatable: to survive a night of unimaginable horror and to process the shattering of her normal, peaceful reality. She is not a fighter, but a regular person forced into extraordinary circumstances.

Her key relationship is with Saya, though it is not one of friendship or deep understanding. Saya, who typically holds humans in contempt, nevertheless saves Makiho's life on multiple occasions, though her specific reasons remain unclear. For Makiho, Saya is a terrifying and enigmatic figure, a savior who is just as frightening as the monsters she hunts. She also has a brief interaction with David, Saya's handler, who stresses the importance of reaching Makiho before the police do, hinting at the government's interest in covering up the incident.

By the end of the film, Makiho has undergone a complete psychological upheaval. After being recovered by David's partner, Louis, she is subsequently seen in an interrogation room, being questioned by government officials about the night's events. However, she finds herself unable to prove her story, as all physical evidence of the battle has been erased, and Saya and David have vanished. In a final, haunting scene, her interviewer shows her a photograph from 1892 featuring a girl who looks identical to Saya, with the word "vampire" written on it.

The story concludes from Makiho's perspective, as she returns to the school. She narrates that she never learned the full truth behind Saya and the creatures she fought and is left to wonder if Saya is still out there fighting, somewhere in the world. She thus represents the lasting, mysterious impact of encountering a hidden, supernatural world, left with only fragmented, unbelievable memories.

Makiho does not possess any notable supernatural abilities or combat skills. Her strengths are entirely human, manifesting as a latent reservoir of courage that allows her to overcome her paralyzing fear for a crucial moment to aid Saya.
