Hiedra Venenosa serves as a supporting antagonist in the anime film's narrative. Transported to feudal Japan alongside other Gotham City villains by Gorilla Grodd's time displacement device, she finds the era's pre-industrial, pollution-free environment deeply appealing.

Upon arrival, she initiates a prolonged two-year military campaign against the displaced Penguin, resulting in a stalemate where neither achieves victory. Her stronghold is a mobile castle, akin to those used by other villainous lords. During their conflict, her castle physically collides with the Penguin's. This clash is abruptly interrupted when Gorilla Grodd, manipulating events, hurtles Two-Face's castle onto them, incapacitating both factions.

Subsequently, Grodd exerts mental control over Hiedra Venenosa and the other subdued villains, compelling them and their forces to construct his combined castle fortress. After freeing herself from Grodd's influence later in the story, she confronts Robin directly and is defeated. Her castle, along with the others, is later absorbed into the massive amalgamated super-robot Lord Joker, formed when the Joker seizes control of Grodd's structure.

Following the villains' defeat and the destruction of the Lord Joker robot, she is returned to the present day alongside the others.
