Dokuro, also known by her full name Dokuro Mitsukai or the nickname Dokuro-chan, is the central character of the series, an angelic being dispatched from a future timeline. Her name is a Japanese homophone for skeleton, a fitting detail given the graphic violence that follows her. Physically, she appears as a young girl with a sweet, high-pitched voice and a deceptively cute appearance that contrasts sharply with her destructive capabilities. She stands 153 centimeters tall and possesses superhuman strength and endurance that she often fails to fully control. Her most distinctive feature is her halo, which is not merely decorative but razor-sharp, functioning with the deadliness of a武士刀, and cannot be safely touched; forcibly removing it causes her severe distress, including uncontrollable diarrhea and comical facial expressions. She is almost always seen carrying her signature weapon, a magical, oversized spiked metal club named Excalibolg.

Dokuro’s personality is a chaotic and volatile mix of childish innocence, manic energy, and brutal impulsivity. She can shift in an instant from being sweet and affectionate to explosive and lethally violent, often in response to minor provocations, misunderstandings, or even her own boredom. While other characters might express displeasure with a shout or a slap, Dokuro expresses hers by drawing her spiked club and gruesomely bludgeoning her victim, who is almost exclusively the series protagonist, Sakura Kusakabe. When she is not killing him, she relentlessly teases him, frequently using sexual innuendo and putting him in embarrassing situations. Despite her destructive tendencies, she is not malicious in a conventional sense; her actions stem from a simplistic, almost childlike lack of foresight and emotional regulation. She speaks with a distinctive verbal tic, ending nearly every sentence with dayo.

Her motivations are rooted in a mission that she ultimately rejects. Originally, Dokuro is a member of an angelic order called Lulutie, sent back in time to assassinate the middle school student Sakura. In the future, Sakura is destined to invent a technology that grants immortality but with a specific side effect: it halts the physical aging of all women at the age of twelve, effectively creating a world catering to his specific preferences, which is deemed an offense to God. However, upon arriving in the past, Dokuro falls in love with Sakura and decides to abandon her deadly orders. Her new goal is to alter the future by keeping Sakura so thoroughly distracted and occupied with the chaos she creates that he will never develop the problematic technology. Ironically, her chaotic and violent method of protecting him often poses a greater and more immediate threat to his life than any other assassin. She lives with Sakura, forcing her way into his home and his life, to personally oversee this mission of distraction and "protection".

Within the story, Dokuro serves as the primary catalyst for all the absurd and violent comedy. Her role is to bludgeon the protagonist to death in increasingly messy and creative ways, only to immediately resurrect him, creating a reset button for the next gag. The core drama of the series revolves around Sakura’s attempts to survive her violent brand of affection while navigating his own feelings for her and his childhood friend. Her most significant relationship is with Sakura, a strange bond defined by her romantic feelings, extreme jealousy, and casual homicide, which she often justifies as a form of expressing her love. She is also rivals and occasional friends with Sabato, another angel assassin sent to complete the mission that Dokuro abandoned, leading to a dynamic of conflict and reluctant camaraderie. She has a younger sister named Zakuro, who appears older and more mature; their relationship is sisterly, though Zakuro sometimes casually suggests that Dokuro should simply finish her mission and kill Sakura.

Throughout the series, Dokuro undergoes very little character development, which is a deliberate choice befitting the show's repetitive comedic structure. Her personality and methods remain consistently erratic from beginning to end. One minor change that occurs is that she grows more comfortable with Sakura seeing her nude as the series progresses. Her core traits, from her violent outbursts to her mission to save Sakura through distraction, remain fixed, ensuring the cyclical nature of the comedy remains intact. Her other activities, such as founding a school club dedicated to watching wood glue dry, which she forces Sakura to join, are simply further expressions of her unchanging, bizarre, and domineering personality.

Her notable abilities are as extreme as her personality. Her primary power is the use of her spiked club, Excalibolg, with which she can kill a person instantly and with great gore. Counterbalancing this destructive power is her most crucial ability: resurrection. By chanting the nonsensical phrase "Pipiru piru piru pipiru pi," she can bring the recently deceased back to life, fully restored and with no lasting harm. She also possesses other magical abilities, such as transforming humans into animals or erasing them from existence entirely, often using these powers for her own convenience or amusement. These abilities, combined with her superhuman strength and her sharp halo, make her an overwhelmingly powerful and unpredictable force within her world.
