Amago Vaccine is a thoroughbred racehorse from the anime Midori no Makibao, recognized as one of the top three horses of his generation alongside the protagonists Midori Makibao and Superhorse Cascade. His full brother is the legendary dual-crown winner Peter II, a horse who was forced into early retirement due to injury. Because of his prestigious bloodline, Amago Vaccine initially struggles under the weight of expectation. He begins his racing career plagued by a lack of self-confidence, suffering defeat after defeat.

Beneath a generally calm and composed exterior, Amago Vaccine possesses a fierce and indomitable fighting spirit. He often appears approachable and friendly, sharing easy conversation with rivals like Makibao. However, when faced with frustration or setbacks, such as being caught in a racing accident caused by another horse, he has been known to secretly vent his anger in private, revealing a fiery passion that lies beneath his cool surface.

Amago Vaccine's primary motivation is to fulfill the dream of his injured brother, Peter II. After Peter II is forced into retirement, he entrusts Amago Vaccine with the goal of winning the Triple Crown, a feat his brother could not achieve. His name is a combination of his owner's name and the word vaccine, a thematic counterpoint to Cascade, whose name evokes a computer virus.

In the story, Amago Vaccine serves as a major rival to Makibao and Cascade, forming a celebrated trio known as the three top horses. His early career is defined by his pursuit of his brother's legacy and his attempts to defeat his powerful rivals. He suffers a significant setback when a fracture, caused by an accident on the track, prevents him from reaching his full potential and forces him to miss the Japanese Derby. He makes a comeback, returning to racing in the same event where his brother was injured. His moment of greatest triumph comes in the Kikuka Sho, a long-distance race on the flat tracks of Kyoto that suits his strengths perfectly. Taking full advantage of his stamina and his familiarity with the course, he finally defeats Makibao to become a G1 winner, achieving his long-held dream.

His key relationships are with his brother Peter II, whose dream he carries; his primary jockey, Yamanaka Mabonosuke; and his two greatest rivals, Makibao and Cascade. Later in his career, he also competes internationally in Dubai and has notable races against other horses like Satomi Amazon and Blitz.

Throughout the series, Amago Vaccine shows significant development. He transforms from an insecure and defeated horse into a confident and triumphant warrior, finally winning a G1 title and earning the respect of his peers.
