De Nisco is perhaps best known for his long-standing association with the anime series Saint Seiya, known in Italy as I Cavalieri dello zodiaco. He is the primary Italian voice of the character Andromeda, also known as Shun, a role he has performed across multiple iterations of the franchise. This includes the original television series, the films such as The Goddess of Discord, The Heated Battle of the Gods, Legend of Crimson Youth, and Legend of Sanctuary, as well as the OVA series The Hades Chapter - Sanctuary, The Hades Chapter - Inferno, and The Hades Chapter - Elysion. His portrayal of this gentle yet powerful Bronze Saint is a defining role of his career.

Beyond Saint Seiya, his anime voice credits are extensive and varied. He voiced the character Sam in the 1985 film The Dagger of Kamui and took on the role of Porthos in the series Anime Sanjushi. He played Arthur in the series Lady Lady!! (also known as Milly, un giorno dopo l'altro) and Steven Martland in the Netflix original anime series Hero Mask. In addition to his work in Japanese animation, De Nisco has a substantial career dubbing characters in Western animation and video games. He has voiced characters like Torpedo in G.I. Joe: A Real American Hero, Mervis and Dunglap in CatDog, and roles in various Beyblade films and series. In the video game industry, he is a prominent voice, lending his talents to major franchises.
