Sakura Yamazaki descends from the Kusamikado lineage, steeped in Japan's spiritual traditions of faith healing. Childhood trauma marked her when her mother died battling the water Aragami Nodzuchi, leading to Sakura being cast out by her village. American government agents subsequently brought her to the United States to train and develop her inherited abilities.

She possesses mastery of Shinto practices, specializing in spells, incantations, protective barriers, and summoning spirit entities, making her a formidable combatant against supernatural threats. Unabashedly narcissistic and supremely confident, she openly touts a self-proclaimed flawless success rate and adopts a flamboyant, attention-seeking demeanor. When not performing duties in traditional robes and glasses, she presents as conventionally attractive and enjoys manipulating romantic interests, particularly to provoke others.

Driven by vengeance against the Aragami responsible for her mother's death, she returns to Japan. Initially operating alone, she pragmatically contracts with the Terrestrial Administration Center (TAC), lending her spiritual capabilities in exchange for payment. While her collaboration is practical, motivations beyond simple extermination hint at unresolved personal stakes.

Sakura exhibits a fierce rivalry with the series' primary protagonist, Momiji Fujimiya, whom she derisively dubs "Sacrifice Girl." This animosity stems partly from Momiji receiving preferential attention, especially from key TAC operative Mamoru Kusanagi. Sakura frequently flirts with Kusanagi to provoke Momiji, reflecting their competitive dynamic in Aragami combat. In the *Blue Seed Beyond* OVA, set two years later, she takes part in U.S. missions against biotechnologically engineered Neo-Aragami, persistently undermining Momiji with overt commentary on physical attributes like breast size.
