Orphaned in Iran at age five when U.S. military forces attacked an ARCAM excavation site, killing his archaeologist parents, Yu Ominae was profoundly traumatized by the sight of their corpses. Relocated to a Japanese orphanage, he befriended Rie Yamabishi before being adopted by relatives in the United States. There, U.S. military operatives abducted him for Project COSMOS, a program brainwashing and training child subjects as elite soldiers. Designated "Number 43," he participated in a South American massacre; executing fleeing civilians triggered memories of his parents' deaths, causing him to slaughter COSMOS personnel in a berserk rage. ARCAM operative Oboro found him and subdued him, facilitating his return to his adoptive family.

Reunited with his uncle Takashi and sister Akiha, Yu grappled with COSMOS-induced violent impulses. During a home robbery, he killed two assailants and nearly killed a third before Akiha intervened, injuring herself. Takashi confronted Yu, urging him to reject being a "murder machine," helping him reclaim his humanity. Yu subsequently traveled with Takashi before joining ARCAM as a Spriggan agent. Under Oboro and knife instructor Bowman, he underwent intensive training to channel his combat skills toward protecting ancient artifacts.

As a Spriggan stationed at ARCAM's Japanese branch, Yu maintained a dual life as a high school student. Frequent mission deployments caused absenteeism and classroom sleeping, leading peers and teachers to view him as a delinquent. He consciously preserved this ordinary school life, seeing it as his anchor to normalcy and a bulwark against reverting to his violent COSMOS persona. He wore a standard gakuran uniform at school and casual attire during downtime. On missions, he utilized an Orichalcum-powered Armored Muscle (A.M.) suit designed by Dr. Mayzel, featuring a tactical vest, power gloves, head protector, bandana, and bandages.

Yu balanced outward optimism and kindness with internal conflict. He valued friendships and loyalty, especially to Rie Yamabishi, whom he protected without revealing his Spriggan role during her artifact-deciphering work for ARCAM. His dedication to ARCAM's mission—sealing dangerous ancient technologies—was unwavering, yet he pledged to destroy the organization if it weaponized artifacts. He resisted unnecessary killing, preferring to incapacitate opponents non-lethally, arguing against executing wounded enemies in Turkey and using fists instead of firearms against Trident soldiers in Romania. This stemmed from his hatred of his COSMOS-instilled brutality, though he accepted lethal force when mission-critical.

Early missions revealed flaws, including overreliance on the A.M. suit's perceived invulnerability and occasional recklessness. Underestimating opponents like the psychic Colonel McDougal nearly proved fatal. He maintained tense interactions with fellow operatives, notably expressing frustration toward Yoshino Somei for complicating missions. His first assignment involved infiltrating a Tokyo research lab to neutralize the Megiddo Flame weapon and its American handler, Major Hummingbat. Later, he escorted Rie Yamabishi to ARCAM and uncovered a shrine on Mount Fuji, reconnecting with her as a childhood friend while withholding his Spriggan identity.

Yu's combat prowess, rated S-Class by ARCAM, combined COSMOS training in firearms, martial arts, and commando tactics with ARCAM enhancements. He wielded weapons including SIG Sauer pistols, knives, and grenades, and piloted a Yamaha V-Max motorcycle for transport. Post-high school, he continued operations, neutralizing the South Pole Shrine's Fire Snakes and Trident forces. He later enrolled in college and undertook missions like securing the Gold Statue of Pachacamac in Mexico while protecting classmate Nanako Kondo. His residence shifted to the penthouse of ARCAM's Japan headquarters, reflecting his enduring role as a top agent.
