She attends Tokiwadai Middle School, earning the title "Tokiwadai Ace" through her high academic standing and formidable powers. Her signature Railgun attack, propelling metal projectiles at three times the speed of sound, inspired her nickname. She has chestnut-brown hair and eyes, consistently wears the Tokiwadai uniform with white slouch socks and brown loafers, and always sports shorts under her skirts.

Daughter of Misaka Tabigake and Misaka Misuzu, Mikoto entered Academy City as a Level 1 esper in elementary school, ascending to Level 5 through intense training. This advancement made her DNA perfect for the covert "Sisters Project," where researchers cloned her under the guise of curing muscular dystrophy. The project generated 20,000 clones, with Accelerator slaughtering 10,000 in the "Level 6 Shift" experiment. Discovering this, Mikoto tried to halt the experiments by destroying facilities and confronting Accelerator directly, ultimately requiring rescue by Kamijou Touma.
