Tadaomi Karasuma is a 28-year-old male officer belonging to the Ministry of Defense's Temporary Special Affairs Division, holding the rank of Lieutenant Colonel. Standing 180 centimeters tall and weighing 85 kilograms, he has a strong, muscular build with spiky black hair and a sharp, penetrating gaze. He was born on August 15. Karasuma is the individual who brought the creature known as Koro-sensei to Class 3-E to serve as their homeroom teacher. His official duties include monitoring Koro-sensei, coordinating with and providing backup for the various assassins sent to the class, and acting as the primary liaison between the students and the government. He holds a valid teaching license and, because Koro-sensei's existence is classified, he is officially recognized as the class's homeroom teacher, though his actual position is that of the assistant homeroom teacher and physical education instructor.

Karasuma’s personality is defined by a strict, professional demeanor that masks a genuinely kind and caring nature. He typically maintains a serious expression and a calm, professional distance from the students, which is a product of his role as a government agent. Despite this, he is deeply committed to the students' welfare and wants them to have a normal school life and a successful future, a desire that often puts him in a difficult position when national orders conflict with the unique environment of Class E. The female students consider him handsome and have described him as cool. He is known for his extreme obliviousness to romance, being completely unaware of his colleague Irina Jelavic's affection for him, a fact that is evident to nearly everyone else around him. This denseness is presented as one of his few personal weaknesses. Even when he becomes aware of her feelings, he prioritizes his work and the mission above all else, believing that romance could interfere with his duties. He likes dogs, but his hidden, intense combat nature causes them to bark at him frantically, and even his smile can exude a powerful intimidation that unnerves trained canines.

Karasuma's core motivation is the successful completion of his mission to assassinate Koro-sensei, which is his top priority as a defense official. However, he is equally driven by his role as an educator. He is determined to see the students of Class 3-E, who are also preparing for high school entrance exams, succeed in life. This leads him to teach them practical assassination techniques under the guise of sports, hoping to provide them with skills that will be useful while ensuring their academic lives are not entirely consumed by the mission. A formative experience was the death of his younger sister, who was weak in both body and spirit. This loss drove him to acquire immense strength to protect anyone, no matter how weak, and to develop a strong sense of impartiality, treating all people equally.

Within the story, Karasuma serves as the crucial adult authority figure and a bridge between the government and the students. He is responsible for bringing in other professional assassins, such as Irina Jelavic and the former trainer Lovro, to aid in the training and mission. He often plays the role of a straight man, reacting with logical bewilderment to the many absurd situations created by Koro-sensei and the students. His combat prowess is legendary within the series, where he is frequently referred to as the strongest man in humanity. He has a distinguished military background, having served in the Ground Self-Defense Force's 1st Airborne Brigade and the Ministry of Defense's Intelligence Headquarters Integrated Intelligence Division. Although his abilities are ineffective against the superhuman Koro-sensei, he possesses top-tier martial arts skills among the human characters. In a hand-to-hand fight against the individual known as the Reaper, who was recognized as the world's strongest assassin, Karasuma won decisively without using any weapons.

Karasuma's key relationships evolve significantly over time. His relationship with the students of Class 3-E begins as a purely professional one, but he gradually becomes a dedicated mentor who invests in their growth, both as assassins and as people. He is particularly influential in recognizing and nurturing Nagisa Shiota's latent talent for assassination. His most prominent relationship is with Irina Jelavic, a professional assassin hired as the class's English teacher. While the students and Koro-sensei orchestrate numerous attempts to bring them together, Karasuma remains frustratingly oblivious to her advances, often misinterpreting them as part of a seduction technique or mission-related activity. Despite his denseness, he shows concern for her safety and, during a critical moment, suggests she leave her life as a killer to work for the Defense Ministry's intelligence division, proposing they live together. His dynamic with Koro-sensei is also notable; he is one of the few people who knows fragments of Koro-sensei's past as a human assassin, and despite referring to him impersonally for most of the series, he calls him by his name during their final farewell.

Throughout the series, Karasuma undergoes significant development. Initially a strict operative focused solely on the mission, he becomes conflicted as his bonds with the students deepen, leading him to question orders that could harm them or their education. When the students decide they want to find a way to save Koro-sensei rather than kill him, Karasuma, after his own period of doubt, supports them as a teacher. However, he also firmly maintains the government's position, clarifying that any plan with even a small risk cannot supersede the national directive to eliminate the threat. In the final stages of the story, when the government detains the students to carry out a laser-based assassination plan, Karasuma covertly provides them with crucial information and expresses his trust in their judgment. Seven years after the events of the main story, Karasuma has been promoted to a director within the Ministry of Defense. He has married Irina, and they have a daughter together.
