Sugita is widely recognized for his distinctive, deep bass voice and his skill in comedic roles, often being referred to as the "first吐槽男" (a title meaning "king of tsukkomi" or straight-man comedy).

His interest in voice acting began in his childhood when he noticed that characters in anime could have voices different from his imagination, yet still be cool. Encouraged by his older brother to pursue a career utilizing his voice, he won an award at a voice actor training school while still in high school and made his debut in 1999. His first leading role came in 2002 as Hideki Motosuwa in the anime series Chobits.

Sugita's career is marked by several iconic and career-defining roles. He is perhaps best known as the voice of the lazy yet powerful samurai Gintoki Sakata in the long-running and beloved series Gintama. Another seminal role is that of the sarcastic and long-suffering Kyon in The Melancholy of Haruhi Suzumiya. His versatility is further demonstrated through roles such as the hot-blooded Joseph Joestar in JoJo's Bizarre Adventure, the boastful yet mighty Escanor in The Seven Deadly Sins, the artistic Yusuke Kitagawa in Persona 5, the strategic Reisi Munakata in the K project, and the powerful Stone Hashira Gyomei Himejima in Demon Slayer: Kimetsu no Yaiba.

Throughout his extensive career, he has voiced a vast number of characters across all genres, including many of those listed by the user. These roles include Kensei Muguruma in Bleach, Almond in I'm the Villainess, So I'm Taming the Final Boss, Pipimi in Pop Team Epic, Gang Manseok in The God of High School, Kōhei Nikaidō in Golden Kamuy, the Lizard Priest in Goblin Slayer, Etsuya Eizan in Food Wars, Tadaomi Karasuma in Assassination Classroom, Hidenori Tabata in Daily Lives of High School Boys, Kaede Manyūda in Kakegurui, Gundam Tanaka in the Danganronpa series, and Marlo Freudenberg in Attack on Titan. He has also lent his voice to films like Dragon Ball Super: Broly as Lemo.

Sugita is known for his close friendships within the industry, most notably with fellow voice actor Yuichi Nakamura. He is also considered the leader of the so-called "Sugita Group," a circle of friends that includes other prominent voice actors. Outside of acting, he is a prolific radio personality, hosting shows like "Anigera! Didooon!!!". His contributions to the industry have been recognized with awards, including the Best Supporting Actor award at the 3rd Seiyu Awards in 2009.
