After training at the Shochiku Music and Dance School, she became a member of the performing group Young 101 and appeared on the NHK music variety show Stage 101 beginning in 1970. Following her time on the show, she worked on the children's program Okaasan to Issho, where her exposure to puppetry and voice actors inspired her to pursue voice acting as a profession.

Ichijō made her debut as a voice actor with the Japanese-language dub of the American television series Man from Atlantis in 1977. She went on to build an extensive filmography in anime, foreign film dubbing, and educational recordings. Beyond anime, Ichijō was highly respected for her dubbing work, most notably as the long-standing Japanese voice of Marge Simpson on The Simpsons. She also dubbed roles for Western films and series, including Beverly Crusher on Star Trek: The Next Generation. Her voice was versatile, allowing her to portray a wide range of characters from mothers and grandmothers to career women and authoritative figures. In 2007, she served as a director of the Japan Actors Union.

Miyuki Ichijō passed away on October 24, 2023, at the age of 76. Her final voice acting credit was in the Detective Conan film released earlier that year, marking the end of a prolific and influential career in the Japanese voice acting industry.
