Mitsuhiko Tsuburaya attends first grade at Teitan Elementary School and co-founded the Detective Boys with classmates Ayumi Yoshida and Genta Kojima following early investigations alongside Conan Edogawa. Raised by teacher parents, he maintains a formal upbringing reflected in his consistent use of the honorific "-san" for elders. His older sister, Asami Tsuburaya, is roughly ten years his senior.
Mitsuhiko employs a scientific, analytical mindset during cases, often citing science books and accumulated knowledge. He proactively prepares for investigations through historical, scientific, and technical study, which aided the group during the Momotaro container riddle in Kurashiki. His practice of recording clues in a Detective Boys notebook—safeguarding against evidence loss—proved vital when original codes were destroyed in a murder case.
Though less physically robust than Genta, Mitsuhiko displays leadership and resourcefulness without Conan or Ai Haibara. Trapped in a cavern during "The Desperate Revival" arc, he deciphered the escape riddle after Conan collapsed. In "Mushrooms, Bears, and the Detective Boys," he fashioned a bandage for an injured Haibara using Conan's technique, carried her to safety, and devised a coded rescue signal. This incident sparked his romantic feelings for Haibara.
His devotion to friends drove him to enter Gunma’s forest alone in "Mitsuhiko's Mystifying Forest" to capture fireflies for Haibara and Ayumi after overhearing their admiration. There, escaped killer Ki'ichiro Numabuchi spared him due to their shared appreciation for fireflies and returned him to his group.
Mitsuhiko feels jealousy when Ayumi shows affection toward Conan but sustains a close bond with Genta despite their mutual romantic interest in Ayumi. In non-canon material, he confided in Ran Mouri about equally liking Ayumi and Haibara, questioning if this made him "a bad person."
He appears with the Detective Boys in the OVA "The Miracle of Excalibur," joining a treasure hunt for a baseball bat planted by Professor Agasa and a coach.
